T4 RNA ligase는 RNA의 5′ phosphate와 3′ hydroxyl 사이에 phosphodiester bond을 형성하도록 촉매 작용을 합니다. 이 효소는 절단된 이중가닥 RNA를 봉합하는 데 가장 활발하지만, 단일가닥 RNA의 말단도 연결할 수 있으며 RNA의 3′ hydroxyl를 DNA의 5´ phosphate에 연결할 수도 있습니다. 효소는 기질의 5’ 말단이 미리 아데닐화(preadenylated) 되어 있지 않은 경우 ATP를 필요로 합니다.
